Artificial breeding techniques

Acquisition and insemination tools

For artificial insemination of earthworms, tools such as a collection cup, a nylon syringe, a nontoxic plastic conduit, and a thermometer are needed. The collection cups are made of non-toxic plastic, the open end is hard, and the bottom is soft and elastic. In this way, the semen can be collected by squeezing the bottom end with a handful of cups, and when the opening of the cup is moved to the male head of the penis, the bottom of the cup is relaxed and a negative pressure is formed to facilitate the inhalation of the semen. Nylon syringes can use medical milliliter syringes. The plastic catheter is made of non-toxic plastic and used with a syringe. As an insemination catheter, it can be replaced and the nozzle can be matched with the syringe head. Thermometers can use ordinary mercury thermometers.

Semen

Semen collection is generally performed once every 2 days to 3 days, and the time should be selected in the afternoon. The semen collected was input into the cloaca of the female nymph within 30 minutes. Semen collection requires two people to cooperate with each other: one is the operator who is responsible for extracting the male ejaculate; the other is the assistant who is responsible for the fixation and collection of semen. Use scissors to cut off the feathers around the male anal and then wipe it with a 65% alcohol swab to avoid disturbing the operation and contaminating the semen. When the sperm is collected, the operator sits in a chair and holds the male's wings in his left hand. He holds his legs on his thighs with his right hand; the assistant's right hand sets a fine cup to collect the semen and the left hand replaces the operator. With his legs and hands. At this time, the operator can stroke the entire palm of the right hand from the back of the male to the back of the plume several times in order to reduce his panic and cause sexual desire. When the male has caused a sexual reflex (exhibiting a tail feather upturn and a genital protrusion rhythmically eversion), the operator loosens his wings, replaces his right hand with his left hand, strokes the tail of the male mantle with his left palm, and the thumb The remaining four fingers were placed on both sides of the cloaca, and the right hand frequently massaged around the cloaca for 5 seconds to 7 seconds in a rapid and dexterous manner to enhance the libido. The operator then squeezed the cloaca with his left thumb and index finger. On the side, under the common extrusion and massage action of the left and right hands, the genitalia is projected to reach a full erection, ie, milky white semen is discharged. At the same time, the assistant accepts semen with a collection cup. Due to the small amount of ejaculate, the close cooperation between the assistant and the operator is very important.

The action during light harvesting should be light, which makes males feel comfortable. This is very beneficial for males and females. Pressing too much can cause excretion of feces and urine, and even make the genitals prominence, the internal capillaries rupture, causing bleeding, thereby contaminating the semen.

Insemination

The produced semen should be diluted approximately 10 times with a warm dilution of 30°C to 40°C. Diluent 0.9% physiological saline can be used, preferably prepared by the following formula: 4.0 grams of sucrose, 1.0 grams of glucose, 1.0 grams of sodium acetate (optimum grade), 0.15 grams of sodium bicarbonate (chemically pure), potassium acetate phosphate 0.2 ml, the pH was adjusted to 7.1, and distilled water was added to 100 ml. The semen produced must be fed within 30 minutes, otherwise it will seriously affect the fertilization rate. Each female was fed 0.1 ml to 0.2 ml of diluted sperm. Each input dose should contain 7500 to 10,000 sperm. Lose 2 times a week, ie 2 consecutive days, once a day.

The insemination work is done by two people. One is responsible for fixing the female and the other is responsible for insemination. During operation, the person responsible for fixing the female scorpion grasps the legs of the female scorpion with his right hand, lifts the female scorpion's double wings with his right limb, and holds his head with his left hand; the person responsible for insemination uses plastic. After taking the semen from the syringe, insert the insemination catheter into the syringe head, insert the right hand index finger into the anus of the female cockroach, and insert the insemination catheter into the cloacal cavity at a depth of 1 cm to 2 cm, and massage it with your fingers. Outside the cloaca, slowly inject the semen. In order to increase the insemination effect, the inseminator can also use the right hand to press hard on the tail of the female scorpion, and use a separate thumb and forefinger to open the anus of the hen and open the fallopian tube mouth (the fallopian tube is located on the left side of the cloaca, The right side of the rectum is opened. The inseminator inserts the insemination catheter obliquely into the fallopian tube by about 1 centimeter, enters the semen, and then restores the anus, completing the insemination.

Be careful not to input air or air bubbles during insemination, otherwise the fertilization rate will be affected. In order to prevent mutual infection, it is best to lose one female sperm for each fertilized egg duct. Since normal water, alcohol, and disinfectants are harmful to sperm, all instruments that come into contact with semen during the insemination operation must not be exposed to these substances. If cleaning is required, they should be washed with a diluent (formula as described above).

Problems that should be noticed in artificial insemination

Elimination of poor males: During the first and second sperm collection training, there will be some inferior males with less genital protrusion, less sperm volume, less sperm density, and poor sperm viability. Marking should be eliminated if it remains so.

Pay attention to semen quality: When collecting semen, only take pure white, high concentration semen, do not take yellow, brown or too thin semen. In order to reduce the pollution of urine, feces, and blood to semen, sperm extraction should be carried out after defecation, and sperm collection should be done lightly. Do not use too much force.

Semen should be handled properly: You cannot dilute the semen with normal water. Diluted semen uses a specially prepared diluent. All instruments that come into contact with semen must not be accompanied by alcohol and disinfectants. The spermatozoa in the range of 10°C to 40°C can maintain a good fertility, especially about 35°C. In the cold season, attention should be paid to the semen temperature, and the temperature of the semen should reach 35°C (a dilution of 35°C to 40°C can be achieved when the semen is diluted). All instruments that come in contact with the semen should use 35°C before the sperm collection and insemination. Flush with ~40°C diluent or saline and allow it to warm up.

Correctly master the insemination technology: insemination should be timely. The spermatozoa are lost within half an hour after sperm collection, otherwise the spermatozoa lose their vigor quickly, which seriously affects the fertilization rate. The amount of insemination should be appropriate. The collected ejaculate was diluted 10 times, and 0.1 ml to 0.2 ml was input per female and a total of 7500 to 10,000 sperm were input.

Insemination time: Usually selected in the afternoon, until most of the females have finished their eggs, preferably in the evening. Insemination usually occurs twice a week (two consecutive days).

Insemination depth: generally 1 cm to 2 cm in the fallopian tube.

Adhere to the health system: the collection of precision cups, syringes, and infusion catheters must be strictly sterilized before they can be used. If these devices are disinfected with alcohol or disinfectants, they must be washed to ensure they are free of these disinfectants, then dried and used for emergency use. If in urgent use, use a semen thinner. The insemination catheter is transfused one at a time, and the used insemination catheter is collected and used for disinfection.
