Scientists at the Massachusetts Institute of Technology and Boston University used biosynthetic methods to engineer the virus, and the modified virus successfully removed biofilms containing harmful bacteria. Bacterial biofilms are bacterial communities that grow on the surface of biological organs or objects. Many human diseases are associated with bacterial biofilms, and bacterial biofilms inside food processing equipment or medical devices can be a source of long-term infection. The surface film of bacterial biofilms is composed of a variety of proteins, polysaccharides and nucleic acids, which can greatly reduce the attack of bacteria by antibiotics. To effectively fight against bacterial biofilms, it must be able to kill bacteria at the same time and destroy the protective film on the surface.
In the experiment, a bacterial-infected virus, a bacteriophage, was used to remove the biobacterial film. They genetically engineered the T7 phage that attacked E. coli to secrete an enzyme that destroys the protective membrane of the surface. The researchers incubated E. coli biofilms on the surface of plastic articles and treated them with modified T7 phage. The results showed that phage killed 99.997% of the bacteria in the biofilm, and the bactericidal ability was 100 times higher than that of the unmodified phage.
According to US regulations on phage application, this result cannot be directly applied to human drugs. It is expected to be used for disinfection of devices in the food and medical industries, as well as for the prevention and treatment of livestock diseases.
