The two most commonly used methods for preserving microbial cultures are cryogenic cryopreservation and liquid paraffin preservation. The first method, cryogenic preservation, is the simplest and most widely applied. It involves storing an agar slant that is free of growth and contamination in a refrigerator at a temperature between 4°C and 6°C. This method requires periodic transfer to fresh slant media every 2 to 3 months to maintain viability. When it's time to use the culture again, it can be re-cultured on new medium and preserved once more.
The second method, liquid paraffin preservation, is particularly useful for long-term storage. In this technique, sterile liquid paraffin is carefully injected into the test tube containing the microbial culture within a laminar flow hood to ensure sterility. The paraffin is added until it covers the surface of the slant by about 1 cm. Afterward, the tube is sealed with a cork or rubber stopper and further secured with wax to prevent evaporation. Once sealed, the tubes should be stored upright in a dry indoor location. This method allows for storage of the culture for over a year without significant loss of viability.
Both methods have their advantages and are often chosen based on the type of microorganism and the available laboratory resources. Cryogenic preservation is ideal for short- to medium-term storage, while liquid paraffin preservation is better suited for longer periods. Proper documentation and regular monitoring are essential to ensure the success of any preservation method.
